Although a group of doctors (Epstein, Barr, & Achong) discovered in 1964 what is now referred to as the 'Epstein Barr Virus' it was not until 1968, when a laboratory technician (who was working with the Epstein Barr Virus) acquired infectious mononucleosis, that the link between the Epstein Barr Virus and mono was established.

Quality Assurance
Each antigen lot is tested for potency using ELISA, and compared to a reference antigen. This ensures the amount of viral protein in each batch is consistent. The total protein concentration reported includes both viral and non-viral proteins, with the ratio dependent on the purity of the antigen.

EBV-VCA

EL-16-03 (lysates) and EL-16-06 (purified VCA) were analyzed by SDS-PAGE and Western blot analysis. (a) immunoblotting EL-16-03 product using a polyclonal antibody to EBV (10 mg antigen), (b) immunoblotting EL-16-03 using a monoclonal antibody to VCA (100 mg), (c) staining EL-16-03 with Coomassie blue (10 mg) and (d) staining EL-16-06 with Coomassie blue (10 mg) and (e) immunoblotting EL-16-06 using a monoclonal antibody to VCA (10 mg).  The Coomassie stain shows that the EL-16-03 lysate (c) is a preparation that contains non-EBV proteins while EL-16-06 purified VCA (d) contains EBV VCA gp125 antigen only.

Inactivation and Safety
Each lot of HSV antigen is inactivated using gamma radiation and this inactivation verified using a sensitive tissue culture based infectivity assay.

P3H3 Cell extract and EBV-VCA gp125 are prepared using detergents that are documented to inactivate any residual viral activity.

Frequently Asked Questions
  1. How can I select which product to use: EL-16-03 or EL-16-06?

Typically, decision whether to use EL-16-03 or EL-16-06 will depend on the application and/or assay design. If an assay is designed to detect antibodies to EBV VCA gp125, then EL-16-06 should be considered first. If the assay is designed to detect antibodies against other EBV proteins, such as EBNA and EA, then EL-16-03 should be considered. Both products can be used for conjugation to solid particles; however, labelling EL-16-06 with detecting moieties (e.g. HRP or antibodies) is likely to give better labelling efficiency due to its high purity.

  1. Can your product be used for coating to solid surfaces?

Yes, both EL-16-03 and EL-16-06 can be used for coating to solid surfaces such as ELISA plates, microparticles (nanogold, latex, magnetic, paramagnetic, etc). EL-16-03 contains low amount of detergent which can be removed if required prior to conjugation. EL-16-06 can be used for conjugation as supplied.

 

  1. How should I handle EBV VCA gp125 antigen?

EL-16-06 should be kept frozen until use. Thawing should be done at 4°C. If entire volume will not be used in an experiment, the product should be aliquoted and frozen to avoid repeated freezing and thawing of the bulk material. On thawing material should be mixed by vortexing. Sonication of EL-16-06 is not recommended.
